摘要 应用合酶链反应(PCR),核酸分子杂交和病毒分离技术检测了7例临床诊断为病毒性脑炎患者及10例其他神经疾病对照组患者的脑脊液(CSF)中单纯疱疹病毒(HSV)DNA。结果PCR检出5例脑炎患者CSF中HSVDNA阳性,其中HSV1型(HSV-1)2例,HSV2型(HSV-2)3例,而核酸杂交方法仅检出2例,其中HSV-1和HSV-2各1例,与病毒分离法结果一致。PCR技术在无菌性脑炎的诊断上具有更高的敏感性和特异性,因而较为实用。实验还比较了2种DNA模板制备方法,煮沸法简便快速,更宜推广。 7 CSF specimens from. clinical definate viral encephalitis patients and 10 from patients with other neurological diseases were examined for herpes simplex virus infection by means of polymerase chain reaction ( PCR) , DNA hybridization and virus isolatioh.Among the viral encephalitis secimens, 5 were shown HSV DHA positive by PCR, 2 of themwere firmed to be HSV-linfection and 3 were HSV-2.Only 2 were shown by DNA hybridization and virus isolation, Result suggested PCR may be more reliable for the diagnosis of viral encephlitis be- cause of its relative higher sensitivity and specificity Two DNA Extraction methods for prepa- ration of PCR DNA template. from CSF had been compared, CSF Boiling method showed more advantages than. Phenol-Chloroform Extraction.
