摘要
目的探讨葛根粗提物(CP)及葛根素(SP)对人小细胞肺癌H446细胞中增殖细胞核抗原(PCNA)、Fas、细胞周期蛋白(Cyclin)D1、细胞周期蛋白依赖激酶(CDK2)、CDK4和p27蛋白表达水平的影响。方法应用免疫组化法检测空白对照组(CO组)、CP组及SP组H446细胞中PCNA、Fas及CDK2表达水平,流式细胞仪检测三组CyclinD1、CDK4和p27蛋白表达水平。结果CP组PCNA、CDK2、CDK4表达水平显著低于CO组(P均<0.05),CyclinD1表达水平显著低于CO组和SP组(P均<0.05),Fas和p27蛋白表达水平显著高于CO组和SP组(P均<0.05);SP组CDK2、CDK4、CyclinD1表达水平显著低于CO组(P均<0.05),Fas和p27蛋白表达水平显著高于CO组(P均(0.05)。结论CP及SP可能抑制H446细胞的增殖,促进其凋亡;CP较SP作用显著。
Objective To investigate the effect of pueraria crude extract(CP) and slandard of puerarin(SP) on protein expression levels of PCNA, Fas, CyclinD1, CDK2 , CDK4 and p27 in H446 cell of human small cell lung cancer. Methods The cells were divded into three groups:the negative control group,the cells treated with CP, the cells treated with SP. Protein expression levels of PCNA, Fas and CDK2 were quantified by immunohistochemistry and those of cyclinD1 ,CDK4 and p27 were quantified by flow cytometry in three groups. Results The protein expression levels of PCNA,CDK2 and CDK4 markedly decreased after treated with CP compared with the negative control group( P 〈 0.05 ). Compared with the negative control group and the group treated with SP ,the protein expression level of Cyclin D1 markedly decreased( P 〈 0. 05 ) after treated with CP and the expression levels of Fas and p27 increased( P 〈 0. 05 ). The protein expression levels of CDK2, CDK4 and CyclinD1 markedly decreased after treated with SP compared with the negative control group ( P 〈 0.05 ) and the expression levels of Fas and p27 increased( P 〈 0.05 ). Conclusion CP and SP probably inhibit growth and induce apoptosis of H446 cells and the effect of CP is stronger than that of SP in the same situation.
