摘要
目的建立梅花鹿鹿茸干组织异种移植动物模型,并观察原始鹿茸干组织在异体继续生长和发育情况。方法将梅花鹿鹿龄为1周岁的初生鹿茸干组织(对照组用前额骨膜)移植到裸鼠头部皮下,观察移植物的生长情况。于移植后60 d切取移植物,同时提取移植物组织的RNA,以cDNA为模板,采用梅花鹿的X型胶原(ColX)的特异性引物进行PCR,检测移植物来源,并进行序列分析。结果实验组受鼠头部均观察到移植物的生长,移植后60 d,移植物平均直径可达8.03 mm,高度可达15.67 mm。PCR产物可见约200 bp的特异条带,测序结果与已报道的梅花鹿的ColX基因的mRNA序列同源性为100%。结论鹿茸干组织移植到裸鼠头部能够继续生长发育,裸鼠可作为研究鹿茸干组织再生发育规律及其调控机制的动物模型。
Objective To develop an animal model of heteroplastic transplantation with dried sika deer antler tissue and observe the further growth of antler as a xenograft. Methods Transplant the dried antler tissue of sika deer aged 1 year to the subcutaneous tissue of head of nude mice, using the prefrontal periosteum of sika deer as control. Observe the growth and development of the graft. Take the graft out 60 d after transplantation and extract the total RNA as a template for synthesis of cDNA. Design a pair of primers according to the ColX sequence of sika deer, and test for the origin of graft by PCR using the synthetic cDNA as a template. Sequence the PCR product. Results The growth of grafts were observed on the head of transplanted nude mice. The mean diameter and height of graft reached 8.03 and 15.67 mm 60 d after transplantation respectively. A specific band at length of about 200 bp was shown on the electrophoretic profile of PCR product. The homology of mRNA sequence of the PCR product to that of ColX gene of sika deer reported was 100%. Conclusion The sika deer antler tissue as a graft further grew on the head of nude mice. Nude mouse may be used as an animal model for study on the pattern and regulatory mechanism of regeneration and development of antler.
