大黄对多发伤合并休克兔小肠上皮细胞超微结构的影响

摘要 目的观察大黄对多发伤合并休克兔小肠上皮细胞超微结构的影响。方法纯种大白兔24只,复制多发性创伤合并休克兔的模型,随机分为三组:治疗组(常规治疗+大黄治疗组),对照组(常规治疗+安慰治疗组),假手术组,每组8只。复苏24h后处死动物,取小肠行电子显微镜检查,并测量细胞线粒体二维平面形态计量学参数和三维平面形态计量学参数。结果电镜下假手术组小肠上皮细胞各结构清晰、完整,微绒毛排列整齐。对照组小肠上皮细胞可见微绒毛倒伏、缺损,异染色质浓缩、边集,线粒体肿胀、嵴模糊甚至空泡化,粗面内质网扩张。大黄治疗组小肠上皮细胞结构基本完整、清晰、微绒毛轻度排列紊乱,散在性脱落,肿胀的线粒体和扩张的内质网少见,细胞间连接清晰可见。与对照组比较,假手术组及治疗组线粒体数较多,周长、面积、最大直径、最小直径及等效直径均较小(P<0.01)。而假手术组与治疗组的上述参数间差异均无显著性(P>0.05)。与对照组比较,假手术组及治疗组体积密度较小(P<0.01),面积密度、粒子数密度、比表面、形状因子较大(P<0.01)。结论大黄能减轻多发伤合并休克后小肠上皮细胞超微结构病变,大黄对多发伤合并休克模型兔小肠损伤有显著保护作用。 Objective To investigate the effect of Rhubarb on the ultrastructure of small intestinal epithelial cell in multiple traumatic rabbits with complicating shock. Methods Twenty four rabbits were randomly divided into 3 groups, 8 in each group, namely sham operation group, treatment with Rhubarb group (treatment group),treatment with normal saline group (control group), multiple traumatic rabbits with complicating shock animal model was established according to the Chen's method. 24 hours after seriously multiple trauma the animals were killed, small intestinal epithelial tissues were obtained for microscopic examination. The two-dimensional structural parameters and three-dimensional structural parameters of mitochondria were calculated. Results obvious pathologic lesions of the organs were seen in control group. But in treatment group, there were less pathologic lesions of the organs, and in sham operation group, there weren't obviously pathologic lesions of the organs. Structural parameters of mitochondria: In control group, there were the least mitochondria and the swelling of mitochondria was severe. In treatment group, the mitochondria were more than that of control group and the swelling were slight. Conclusions Rhubarb can obviously protect small intestine from seriouslymultiple traumatic rabbits with complicating shock.
