JNK通路在氯通道阻断剂抑制Aβ25-35诱导细胞凋亡中的作用

摘要 目的探讨JNK通路在氯通道阻断剂4,4'-diisothiocyano-2,2'-disulfonie acid stilbene(DIDS)及Phloretin在β淀粉样蛋白活性片段(Aβ,Aβ_(25-35)诱导PC12细胞凋亡过程中作用。方法采用MTT比色法、乳酸脱氢酶(lactate dehydrogen,LDH)活力检测、琼脂糖凝胶电泳法,比较正常对照组、Aβ_(25-35)损伤组和Aβ_(25-35)+Phloretin组、Aβ_(25-35)+DIDS组的PC12细胞存活与凋亡;Westem印迹法检测4组的JNK、P-JNK蛋白表达。结果10μmol/L Phloretin和50μmoL/L DIDS均能抑制Aβ_(25-35)诱导的PC12细胞凋亡,提高细胞存活率,减少LDH释放,降低P-JNK蛋白的水平。结论JNK的磷酸化水平下调,参与了氯通道阻断剂抑制的Aβ_(25-35)诱导的PC12细胞凋亡。 Objective To investigate the effect of JNK transduction pathway on Aβ25-35 induced and chloride channel blockers 4, 4'-diisothiocyano-2, 2'-disulfonic acid stilbene (DIDS) and Phloretin blocked PC12 cell apoptosis. Methods PC12 cell survival and apoptosis rate in control, Aβ25-35, Aβ25-35 + phloretin and Aβ25-35 + DIDS groups were compared by MTY assay, released LDH assay, agarose gel erectrophoresis of DNA. The expressions of JNK and phosphorylated (P)-JNK protein were measured by Western blot assay. Results 10 μmol/L Phloretin or 50 μmol/L DIDS inhibited the apoptosis of PC12 cells induced by Aβ25-35 significantly, elevated cell survival rate, decreased release of LDH, and lowered P-JNK expressions (P 〈 0. 01 ). Conclusions The suppression of P-JNK is involved in Aβ25-35 induced and chloride channel blockers blocked PC12 cell apoptosis.
