目的:观察SSB多肽与DNA抗原单独和共同免疫动物时抗体产生的特征。方法:设计合成SSB214 ̄225aa多肽。用SSB多肽、双链(ds)-DNA、SSB+DNA、血蓝蛋白及PBS分别免疫新西兰白兔,用ELISA法检测SSB多肽抗体,免疫荧光法检测ds-DNA抗体,免疫印迹法检测可提取核抗原(ENA)抗体。结果:ELISA检测显示SSB多肽免疫兔能诱导相应抗体产生,联合免疫组SSB抗体的滴度高于单独SSB免疫组。免疫荧光法检测显示联合免疫组及SSB免疫组各有1只兔产生抗ds-DNA抗体,免疫印迹法检测显示联合免疫组2只兔除产生了抗SSB抗体外,还产生了抗RNP、抗Sm抗体,提示产生表位扩展。结论:合成SSB多肽与DNA共同免疫兔后产生的抗SSB多肽抗体滴度高于单独SSB免疫组,并更易产生表位扩展。
Objective: To observe profile of antibody production in rabbits immunized with synthesized SSB peptides alone or with SSB plus dsDNA. Methods: 214-225 aa peptide of SSB antigen was synthesized. Rabbits were immunized with synthetic SSB peptides linked with keyhole limpet hemocyanin; dsDNA; SSB plus DNA; KLH and phosphate buffered solution (PBS). SSB peptide antibody was measured by ELISA, ENA antibody and dsDNA antibody were measured by immunoblot and immunofluorescence respectively. Results: Specific anti-SSB peptide antibody was produced after immunization of rabbits with SSB peptide alone or SSB plus dsDNA. The level of SSB peptide antibodies in co-immunization group was higher than that of SSB immunization group. Antibodies against RNP, Sm and/or dsDNA were found in rabbit of co-immunization group. In addition, antibody to dsDNA was also found in rabbits immunized with SSB peptide, suggestive of epitope spreading. Conclusion: Epitope spreading was observed in immunized rabbits especially in those immunized with SSB and dsDNA.
