脑创伤早期血清差异蛋白筛选与诊断 被引量:3

Screening of serum biomarkers and establishment of diagnostic fingerprint in early severe traumatic brain injury patients

摘要 目的应用表面加强激光解吸离子化飞行时间质谱(surface enhanced lase rdesorptionionization time-of-fight mass spectrometry,SELDI-TOF-MS)技术筛选重型颅脑创伤(severe traumatic brain injury,sTBI)患者早期血清差异表达蛋白,初步建立sTBI患者早期血清蛋白诊断指纹图谱。方法获取临床收住院早期未接受药物干预治疗的重型颅脑创伤患者(GCS3~8分)与健康体检者血清样本,根据从创伤到入住院时间间隔将病人血清分为≤1h、1-3h和〉3-8h组,分别混合各组血清样本取消个体差异,利用弱阳离子芯片CM10经质谱分析血清中差异表达蛋白,根据检测结果初步分析建立sTBI病人早期血清蛋白诊断指纹图谱。结果样品血清中共获得71个有效蛋白质峰,有7个峰与对照组比较有差异,其中m/z4210、m/z7116和m/z7320在3组病人中均显著升高(P〈0.05);m/z4977和m/z8719在3组中均显著降低(P〈0.05);m/z4679的升高出现在1-3h和〉3-8h组;m/z9431的升高只出现〉3-8h组。结论应用SELDI-TOF-MS技术可寻找到多种血清差异蛋白表达,这些蛋白可能参与sTBI患者早期分子病理事件,它们的发现将会为临床药物筛选和(或)新药物开发提供重要参考。 Objective To seek serum biomarkers and establish serum protein fingerprint in early severe traumatic brain injury patients by surface enhanced laser desorption-ionization time-of-fight mass spectrometry. Methods Serum samples were obtained from 86 cases of early untreated traumatic brain injury patients( GCS scores of 3 to 8) and 50 cases healthy people. Three groups ( ≤ 1h, 1-3h and 〉 3-8h) were determined by the intervals from injury occur to admission. Serum samples were admixed to eliminate individual variation. Protein chip CM10 and SELDI-TOF-MS were used to detect serum differential expression proteins, and establish diagnostic fingerprint of early traumatic brain injury patients. Results 71 protein peaks were detected in serum samples, m/z 4210, m/z 7116 and m/z 7320 in all three groups were significantly higher than control group, but m/z 4977 and m/z 8719 were lower, m/z 4679 was increased in 1-3h and 〉 3-8h groups, and m/z 9431 increased only in 〉 3-8h group. Conclusion Some differential expression proteins can be found by SELDI-TOF-MS. These proteins maybe involve in early pathologic molecular events, and direct clinical drug screening and/or new drug development.
