摘要
目的 观察大鼠胰腺上皮细胞来源类胰岛样细胞簇的胰岛素分泌功能及移植至糖尿病模型大鼠的效果。方法 将传代培养6个月以上的大鼠胰腺上皮细胞经高糖、10mmol/L尼克酰胺、10μg/L肝细胞生长因子和10μg/L成纤维细胞生长因子分化诱导后,分化成类胰岛样细胞簇,应用双硫腙染色法进行鉴定;应用放射免疫法检测培养液中胰岛素和C肽分泌水平;收集成熟的类胰岛样细胞簇,移植到糖尿病鼠的肾被膜下,检测糖尿病鼠血糖水平。结果诱导后1周,自单层的扁平上皮细胞以出芽的方式生长出三维的类胰岛样细胞簇,双硫腙染色后呈桔红色。随着类胰岛样细胞簇的成熟和数量的增加,培养液中的胰岛素水平逐渐升高,并对高糖刺激有反应性。移植实验表明大鼠胰腺上皮细胞来源的类胰岛样细胞簇移植可以将糖尿病鼠的血糖从(22.43±0.23)mmol/L^-1(28.96±1.52)mmol/L逆转至(9.60±0.34)mmol/L^-1(11.80±0.98)mmol/L之间。结论大鼠胰腺上皮细胞来源的类胰岛样细胞簇具有胰岛素分泌功能,移植后可以逆转糖尿病大鼠的高血糖状态。
Objective To observe the secretory function of the islet-like cells derived from the pancreatic epithelial cells (IPECs)and the effect of IPEC allograft transplantation on diabetic rats. Methods The pancreatic epithelial cells had been subcuhured for 6 month Induction differentiation of IPECs was performed by the addition of high glucose (25.5 mmol/L), nicotinamide (10 mmol/L), hepatocyte growth factor (10 μg/L) and basic fibroblast growth factor ( 10 μg/L). IPECs were identified by dithizone (DTZ) staining. Insulin and C peptide contents in culture medium were measured by radioimmunoassay. These IPECs were harvested and transplanted into the renal subcapsular region of the diabetic rats, and then their blood glucose level was monitored every 2 days after transplantation. Results Cultures were followed for the appearance of round cells budding upward from the epithelial monolayers 1 week after induction differentiation. IPECs were stained nacarat with DTZ. The levels of insulin in medium were also increased with the maturing and the increasing quantity of the IPECs. The IPECs was responsive to high glucose stimulation. The renal subcapsular transplantation of IPECs in streptozotocin-induced diabetic rats reversed hyperglycaemia from (22.43 ±0. 23 ) mmol/L-( 28.96 ±1.52 ) mmol/L to (9.60 ± 0. 34 ) mmol/L-( 11.80 ± 0. 98 ) mmol/L. Conclusion IPECs can secrete insulin and correct hyperglycaemia in the diabetic rats.
