摘要
目的研究中药九节茶提取物对小鼠免疫性肝炎和急性炎症的作用,并且通过其对大鼠腹腔中性粒细胞体外释放白三烯的影响,探讨可能的作用机制。方法通过建立痤疮丙酸杆菌和脂多糖(P.acnes-LPS)诱发小鼠肝损伤模型,用酶标仪测定小鼠血浆丙氨酸氨基转移酶(ALT)活性水平,利用反相高效液相法(RP-HPLC)测定大鼠腹腔中性粒细胞释放白三烯B4(LTB4)含量。观察九节茶提取物对2,4-二硝基氟苯(DNFB)诱发变应性接触性皮炎(ACD)的影响,对巴豆油致小鼠急性耳肿胀及对角叉菜胶引起小鼠足肿胀的影响,评价九节茶提取物对迟发性过敏反应及急性炎症的作用。结果与模型组相比,中药九节茶提取物125、250及500mg·kg-1均能抑制由P.acnes-LPS诱发小鼠血浆ALT活性的升高,其抑制率分别为55.1%、70.3%及78.5%,并对大鼠腹腔中性粒细胞体外释放LTB4呈剂量依赖性抑制作用。此外,九节茶提取物对DNFB诱发ACD、巴豆油致小鼠耳廓肿胀及角叉菜胶引起足肿胀也显示一定程度的抑制作用。结论中药九节茶提取物对于P.acnes-LPS引起的免疫性肝炎有较好的抑制作用,其作用机制可能与抑制中性粒细胞释放白三烯相关;而对于急性炎症也有一定的作用。
Aim To investigate the effects of Sarcandra glabra (Thunb) Nakai extract ( SGE ) on mice model of immunological hepatitis and acute inflammation, and its influence on release of leukotriene B4 from rat peritoneal neutrophil in vitro. Methods The immunological liver injury model was established by injection of Propionibacterium acnes and lipopolysaccharide (P. aces- LPS) via lateral tail vein. Hepatic function was evaluated by assessing alanine aminotransferase level (ALT) in plasma. HPLC methods were performed to detect the contents of leukotriene B4 ( LTB4 ) released from murine peritoneal neutrophil in vitro. The effect of SGE on allergic contact dermatitis (ACD) induced by DNFB was studied in micerto observe its effect on delayed-typed-hypersensitive(DTH). Croton oil and Carrageenan were used as well to observe its effect on acute inflammation. Results Compared to the model group, 500,250 and 125 mg · kg^-1 SGE could inhibit the activities of ALT in mice plasma, and the inhibitory rate was 78.5%, 70. 3% and 55. 1% respectively. SGE also inhibited the release of leukotriene B4 in murine peritoneal neutrophil in vitro, in a dose-dependent manner. Meanwhile, SGE could ameliorate the ear swelling in ACD model and acute inflammatory model induced by croton oil. It reduced the thickness of the mice hind paw induced by carrageenan as well. Howev-er, none of them was in significantly dose-dependent manner. Conclusion The results indicated that SGE exerted potential effects on immunological hepatitis and the mechanisms might be partly related to its inhibitory effect on release of LTB4 from neutrophil. SGE also showed partial effect on acute inflammation. leukotriene B4.
