摘要
目的:建立一种在光动力学治疗中癌细胞内光敏剂含量的检测方法。方法:采用RF-540型荧光分光度计,最大荧光激发波长394nm;发射波长610nm,将癌细胞破碎后释放出的光敏剂,进行测量,再根据标准曲线计算细胞内光敏剂含量,同时测定培养液中光敏剂含量。结果:该方法准确可靠,误差小于原液光敏剂浓度的7.5%;培养的SW480细胞内光敏剂浓度在加药后8小时和16小时达到较高浓度,有显著意义,再延长时间,细胞内光敏剂含量增加不显著。结论:该方法准确、可靠、简便,有重要意义。
Objective:To establish a detection method for the intracellular concentration of photosensitizer in photodynamic therapy. Methods: A model of RF-540 fluorescence spectrophotometer was used to detect the intracellular photosensitizer of human colon carcinoma cells after crushing the cell, then the concentrations were calculated by standard curve of PSD.Results: The method was proved of accuracy and reliability, with error less than 7.5 percent of the original PSD concentration of culture solution. In 8,16 hours after adding of the drug, intracellular PSD levels increased significantly, but no significant difference of PSD levels in more than 16 hours or 24 hours. Conclusion: This is an accurate and trustworthy method in detection of intracellulor concentration of photosensitizer.
