摘要
目的探讨细胞因子致鼠小肠移植中小肠缺血再灌注损伤中的机制及丹参的保护作用。方法将60只大鼠随机分为假手术组、模型组和丹参治疗低、中、高3个剂量组,通过夹闭SAM模拟大鼠小肠移植中小肠缺血再灌注模型。ELISA法检测再灌注2h后各组肠组织与血浆中TNF-α、IL-1β和IL-8的含量及肠黏膜损伤程度。结果模型组血浆及肠组织中TNF-α、IL-1β、IL-8含量较假手术组明显升高(P<0.01),肠黏膜损害明显(P<0.01);治疗组上述细胞因子水平较模型组下降(P<0.01),且随丹参剂量增加呈逐渐下降趋势,肠黏膜损害减轻(P<0.01)。结论丹参对小肠移植中小肠缺血再灌注损伤具有一定的保护作用,其机制可能与抑制炎症细胞因子的表达有关。
Objective To explore the protective effect of Salvia mihiorrhiza on small intestinal ischemia reperfusion injury during intestinal transplantation induced by cytokines in rats. Methods Sixty Sprague Dawley rats were randomly divided into sham operation group, model group and low, mid, high doze treatment group, the superior mesenteric artery was occluded and then released to simulate the model of small intestinal ischemia reperfusion during intestinal transplantation. Tumor necrosis factor -α, interleukin - 1β, interleukin - 8 both in the plasm and small intestinal tissue were evaluated by ELISA 2 hours after reperfusion, changes of pathology in intestinal mucosa were observed. Results TNF - α, IL - 1β, IL - 8 were significantly higer( P 〈 0.01 ) in the plasm and small intestinal tissue in model group than that in sham operation group, intestinal mucosa was obviously injured( P 〈 0.01 ) ; cytokines in treatment group were lower than that in model group( P 〈 0.01 ), and a negtive correlation was found between the dose of Salvia mihiorrhiza and content of cytokines, intestinal mucosa injury was alleviated( P 〈 0.01 ). Conclusions Salvia mihiorrhiza has some protective effect on intestine ischemia reperfusion injury during intestinal transplantation, the probable mechanism is related to inhibit the expression of inflammatory cytokines.
