摘要
活化素受体样激酶(Activin receptor-like kinases 6,ALK6)是转移生长因子β超家族成员的受体,具有介导细胞发育和分化等重要功能。以人的ALK6蛋白结构域分析为依据,设计了6对特异性引物,再以贵州白山羊血液基因组DNA为模板,扩增出ALK6基因外显子5至外显子10相应的DNA片段。对6个外显子进行SSCP和RFLP分析,结果表明:ALK6基因外显子5至外显子10中没有发现单碱基突变,说明在绵羊、人等ALK6基因中发现的碱基突变在贵州白山羊中的自然发生率极低,该基因在贵州白山羊群体中很保守,其保守性对山羊的发育调控具有重要的意义。
The activin receptor-like kinases 6 (ALK6) is a receptor of the transforming growth factor-β (TGF-β) family members, which regulates the cell growth and differentiation. Six pairs of specific primers were designed according to the structure analysis of ALK6 in human to amplify the corresponding DNA sections from No. 5-10 exons of ALK6 gene by taking the genomic DNA as templates extracted from 277 blood samples of Guizhou white goat. The results from analyzing six exons by SSCP and RELP show that the mutation of the single base in No. 5-10 exons of ALK6 is not detected, which means ALK6 gene in Guizhou white goat is very conservation which has the important significance for regulating and controlling development and differentiation of goat.
