摘要
目的研究沙利度胺对胶原诱导型关节炎大鼠的关节炎症及血浆血管内皮生长因子(VEGF)表达的影响。方法采用胶原诱导型关节炎(CIA)大鼠为动物模型,经给予沙利度胺治疗,测定不同时间点大鼠的足爪厚度、血浆VEGF浓度,进行踝关节病理评分和放射学检查,并与正常对照组、单纯造模组、甲氨蝶呤治疗组相比较。结果沙利度胺可以有效减轻CIA大鼠足爪肿胀程度,降低血浆VEGF浓度,疗效与甲氨蝶呤相似。结论沙利度胺可以通过影响VEGF的表达,抑制滑膜炎症及周围组织破坏,有效减轻CIA大鼠关节的炎症程度。
Objective To determine the effects of Thalidomide on arthropathy and in collagen Ⅱ -induced arthritis (CIA) rat models. Methods A group of CIA rat models were treated with thalidomide. Of this group, the thickness of rat claw and plasma VEGF concentration, pathological score and radiography of ankle joint were evaluated at different time spots and compared with those of normal group, untreated CIA group and MTX-treated group. Results Thalidomide was showed to result in relieved swelling of ankle joint, reduced plasma concentration of VEGF, lowered pathological score, and delayed progression of bony destruction, with therapeutic effects comparable to MTX. Conclusion Thalidomide may effectively ameliorate the arthropathy in CIA rat models by modulated VEGF expression, suppressed synovitis and bony destruction.
