摘要
将30日龄四川白鹅分别肌肉注射接种50μg、100μg和200μg的GPV-VP3基因疫苗(pcDNA-GPV-VP3),以生理盐水和空载体质粒pcDNA3.1(+)为对照,于免疫接种后1h、12h、1d、3d、7d、21d、35d、63d、105d和217d采集全血以及各组织器官,用实时荧光定量PCR(FQ-PCR)检测pcDNA-GPV-VP3在雏鹅体内的动态分布。结果表明:①pcDNA-GPV-VP3各剂量免疫雏鹅1h即可在各组织中被检测到,其中注射部位含量最高,肝、淋巴器官(脾、法氏囊、胸腺、哈氏腺)含量较高;②到217d时,50μg组免疫雏鹅各个组织器官内仍检测到pcDNA-GPV-VP3的存在,但多数组织器官中的含量比1h时约少了103~104;③血液中pcDNA-GPV-VP3的含量较少,且1h~217d各时间点的差异不显著(P≥0.05);④不同剂量pcDNA-GPV-VP3免疫雏鹅各组织中的含量呈现的总体规律为200μg组>100μg组>50μg组,但差异不显著(P≥0.05)。因此,pcDNA-GPV-VP3免疫雏鹅后1h时可分布至雏鹅体内各组织器官中并持续存在217d以上。
This paper concentrates on 30 - day - old geese were administered with 50μg, 100μg, 200μg of pcDNA - GPV - VP3 respectively by the route of intramuscular injection with Normal saline, pcDNA3.1 ( + ) as control. To detect the recombinant plasmid in the samples of peripheral blood and inoculated muscle, heart, liver, spleen, lung, kidney, brain, thymus, gland, harderian gland, bursa of fabricius, pancreas, duodenum, jejunum, ileum, rectum, appendices and serum with FQ- PCR at the interval of 1 h, 12 h, 1 d, 3 d, 7 d, 21 d, 35 d, 63 d, 105 d and 217 d after administration. Results showed that:①The pcDNA- GPV- VP3 could be detected in all tissues and peripheral blood at 1 h past infection. The copy number was the cargese at the skin of injeeted spat, then come liver, and lymphatic organ (spleen, fabricius, thymus, gland of harder); ②At 217 d post administration, pcDNA-GPV- VP3 could still be detected in tissues of 50 peg dose group. But the copy number decreased about 10^3 ~ 10^4 copies in the most tissues than it was at 1 h post administration; ③The changement of the copy numbers in the serum was always small and not obvious with the time (P≥0.05) ; ④The quantity of pcDNA- GPV- VP3deteeted in the experimental groups showed the relations with the quantity of the inoculation doses, but not proportion (P ≥0.05). The results demonstrated that pcDNA- GPV- VP3 could distribute in all tissues of geese at at 1 h post infection and existed in those after 217 d post infection.
