摘要
目的 研究胰岛素样生长因子结合蛋白相关蛋白1(insulin-like growth factor binding proteinrelated protein-1,IGFBP-rP1)在转化生长因子β1(tronstorming growth factor β1,TGFβ1)诱导的肝星状细胞中表达的变化及抗IGFBP-rP1抗体对TGFβ1,诱导的肝星状细胞产生Ⅰ型胶原的影响。方法大鼠肝星状细胞株(HSC-T6)体外培养,分别设空白对照组(加入等量PBS)、不同浓度的TGFβ1,处理组、不同浓度的抗IGFBP-rP1抗体处理组,处理因素作用24h,采用免疫组织化学染色、Western blot检测IGFBP-rP1在HSC-T6中的表达,ELISA检测Ⅰ型胶原的含量并进行IGFBP-rP1与Ⅰ型胶原的相关性分析。结果1 TGFβ1,各处理组IGFBP-rP1的表达均较空白对照组显著增强。抗IGFBP-rP1抗体可以拮抗TGFβ1,诱导的HSC-T6产生的过多Ⅰ型胶原。IGFBP-rP1与Ⅰ型胶原呈正相关(r=0.833,P〈0.01)。结论 IGFBP-rP1参与了肝纤维化的形成,且可能在肝纤维化的发生、发展中占有重要地位。
Objective To study the expression of insulin-like growth factor binding protein-related protein-Ⅰ (IGFBP-rP1) in TGFβ1 induced hepatic stellate cell and the effects of anti-ⅠGFBP-rP1 antibody on the collagen Ⅰ production of TGFβ1 induced hepatic stellate cell. Methods Rat hepatic stellate cell line (HSC-T6) was cultured in vitro , and then was incubated with phosphate buffered solution ( blank control group), TGFβ1 at 2 μg/L or 4 μg/L (TGFβ1 group), anti-ⅠGFBP-rP1 antibody at 0.1 μg/L, 1 μg/L or 2 μg/L (anti-ⅠGFBP-rP1 antibody group) or both (TGFβ1 + anti-ⅠGFBP-rP1 antibody group). Twenty-four hours later, the expression of IGFBP-rP1 in HSC-T6 was detected by immunohistochemical staining and western blotting; the content of collagen Ⅰ in culture supernatant of HSC-T6 was measured by ELISA; the relationship between IGFBP-rP1 and collagen I was analyzed. Results The expression of IGFBP-rP1 in TGFβ1 group was significantly higher than that in blank control group. The result of ELISA showed that anti-ⅠGFBP-rP1 antibody antagonized the over-production of collagen I by TGFβ1 induced HSC-T6. There was a positive correlation between IGFBP-rP1 and collagen Ⅰ ( r = 0.833,p 〈0.01 ). Conclusions IGFBP-rP1 may play an important role in the formation and development of liver fibrosis.
