摘要
目的观察肺动脉内皮细胞血小板源性生长因子(PDGF)旁分泌及其调节在缺氧性肺动脉高压中所起的作用。方法采用核酸分子杂交技术,分析缺氧时小牛肺动脉内皮细胞(PAEC)表达PDGF-A链、-B链mRNA的变化及外源一氧化氮(NO)对其表达的调节作用。结果缺氧(2.5%O2和0%O2)早期(1.5,3h)能非常显著地增加肺动脉内皮细胞PDGF-A,PDGF-B链mRNA的表达(P<0.001,vs常氧组);缺氧6~48h与常氧组相比PDGF-A链无明显差别,PDGF-B链mRNA表达增加(P<0.05)。外源NO能非常显著地降低常氧肺动脉内皮细胞PDGF-A链(P<0.01)和常氧PDGF-B链(P<0.001)mRNA的表达水平,而不能明显地抑制缺氧PDGF-A,-B链的表达。结论缺氧可能一方面通过刺激肺动脉内皮细胞PDGF基因表达加强,一方面减弱NO对PDGF基因表达的抑制作用,从而参与缺氧性肺动脉高压的肺血管收缩反应加强和肺血管结构改建的发生过程。
Aim\ To investigate the role of platelet derived growth factor(PDGF) of pulmonary arterial endothelial cells(PAEC) under hypoxic condition may play important roles in the development of hypoxic pulmonary artery hypertension(HPAH).\ Methods\ The effects of hypoxia on PDGF gene expression in new bovine PAEC and the modulation by nitric oxide(NO) were measured by dot blot hybridization.\ Result\ PDGF A, B chain mRNA expression increased significantly when PAECs were incubated under 0% and 2 5% O 2 hypoxic condition for 1 5 ̄3 h( P<0 01, vs 21% O 2 control).\ PDGF B chain mRNA kept on high level till 48 h( P < 0 05), whereas, PDGF A chain mRNA did not increase significantly during 6 ̄48 h.\ Exogeneous NO donor(SIN 1) significantly inhibited PDGF A chain( P <0 01) and PDGF B chain( P <0 001) mRNA expression under normoxic condition(21%O 2), but not inhibit under hypoxic condition(2 5% O 2 and 0% O 2 ).\ Conclusion\ These results show that hypoxia may stimulate the expression of PDGF in PAEC in vitro and the inhibition of PDGF mRNA expression by NO under 21%O 2 condition is eliminated by hypoxia, which may lead to the development of HPAH.
