摘要
目的:建立测定雷贝拉唑钠血药浓度的高效液相色谱法。方法:采用Hypersil C18柱(4.6mm×200mm,5μm),以乙腈-水(32∶68)为流动相,流速1.0ml/min,检测波长288nm。结果:本方法线性范围5~1600μg/L,r=0.9993,回收率为95.48%~99.13%。日内及日间RSD均小于4%。结论:本方法灵敏、准确、重现性好,适用于雷贝拉唑钠药代动力学研究。
Objective:To set up a reverse phase HPLC method for determining the concentration of rabeprazole sodium in serum in order to study its clinical pharmacokinetics.Methods:The chromatographic separation was carried out on Hypersil C18 column(4.6 mm×200 mm,5 μm), the mobile phase was acetonitrile-water(32:68) and flow rate was 1.0 ml/min. The detector wavelength was 288 nm.Results:The linear range was 5-1 600 μg/L, r=0.999 3. The method recovery was within 95.48%-99,13%,within-day and between-day R SD were less than 4%.Conclusion :The method is sensitive, specific and reproducible for the determination of rabeprazole sodium concentration in humam plasma.It is very suitable for pharmacokinetics study of rabeprazole sodium.
