摘要
目的研究血液代用品聚乙二醇修饰猪血红蛋白(PEG-pHb)的抗原性。方法通过计算机模拟,比较天然人、兔和猪血红蛋白之间氨基酸序列、空间构象和抗原表位的差异。将PEG-pHb按兔血量的10%输入兔耳缘静脉,免疫多次,制备抗体;将pHb与PEG-pHb分别与免疫佐剂混合,按常规方法免疫家兔,制备抗体。用免疫印迹法与ELISA法检测抗原与抗体之间的交叉反应。结果天然猪血红蛋白与人血红蛋白的分子构象几乎一致,二者有高度相似的抗原性;PEG-pHb溶液反复大量输入家兔体内未检测出抗体。结论PEG-pHb可有效地降低pHb的抗原性。
OBJECTIVE To investigate the antigenicity of PEG-pHb as blood substitute. METHODS The differences of the amino acids sequences, stereo-structures and antigenic epitopes among natural porcine Hb, natural human Hb and natural rabbit Hb were compared by computer simulation. PEG-pHb solution was infused into the vein of rabbits for several times and the antibody was obtained. PEG-pHb and natural pHb were mixed with freund's adjuvant respectively and the rabbits were immunilized by general methods. The antigenic activity of PEG-pHb and natural pHb was studied by the means of immunoblotting assay and ELISA. RESULTS The configurations of natural porcine Hb and human Hb were almost the same, and so do the antigenicities. No antibody and immunological reactions were found in rabbits after infusion of PEG-pHb. CONCLUSION PEG-pHb reduced the antigenicity of pHb effectively.
