摘要
以棉枯萎镰刀菌菌丝体加孢子提取的蛋白质为抗原免疫家兔制备的2种抗血清As2、As10中成分复杂,用琼脂双扩散法和比较对流免疫电泳法测定特异性,交叉反应明显。As2用木贼镰刀菌的异种抗原、As10用禾谷镰刀菌的异种抗原吸收排除非特异性成分后,抗血清的专化性提高,消除了交叉反应。特异性测定结果表明:利用此种经吸收后的抗血清检测棉枯萎镰刀菌可以准确地鉴定到尖孢镰刀菌种的水平,但不能区分专化型。
In this study, the soluble proteins used as antigen were extracted from the mycellia and conidia of the pathogen Fusarium oxysporum f. sp. vasinfectum of cotton fusarium wilt, and the antisera (As2, As10) obtained by immunizing the rabbits contained complicated composition. The nonspecific reaction affected the serodedected result by agar dimension diffusion (ADD) and comparative immunoosmopheresis (CIOP). By mixing As2 and the antigen Fusarium equiseti , As10 and the antigen Fusarium graminearum , then eliminating the nonspecific composition, the specificity of antisera was improved. The results of this study revealed that As2 and As10 can detect the species of Fusarium oxysprorum , but can′t detect forme species.
