摘要
目的:观察预先给予ifenprodil是否可以预防坐骨神经慢性压榨性损伤(CCI)大鼠的神经病理性疼痛。方法:体重在180~200g的雄性SD大鼠随机分为4组:假手术+生理盐水组、假手术+ifenprodil组、CCI+生理盐水组及CCI+ifenprodil组。建立外周神经慢性压榨性损伤(CCI)动物模型,假手术组大鼠同样分离坐骨神经,但不结扎神经。假手术+ifenprodil组及CCI+ifenprodil组大鼠在术前30min及术后第1、2d连续三天腹腔内注射ifenprodil(10mg/kg);假手术+生理盐水组及CCI+生理盐水组大鼠则以同体积生理盐水代替ifenprodil进行腹腔内注射。使用vonFrey纤维测量大鼠手术侧机械痛阈。比较各组大鼠手术前后的机械痛阈。结果:CCI+生理盐水组大鼠的机械痛阈在术后第7d及14d均明显低于术前(P<0.05);其余各组大鼠的机械痛阈在术后较术前无显著性差异(P>0.05);CCI+ifenprodil组大鼠术后7d及14d的机械痛阈显著高于CCI+生理盐水组(P<0.05)。结论:预先给予选择性作用于NR2B亚基的NMDA受体拮抗剂(ifen-prodil)可有效预防外周神经损伤所导致的神经病理性疼痛。因此NR2B的活化在神经病理性疼痛的形成过程中起重要作用。
Objective : The present experiment investigated if the pain behavior of chronic constrictive injury (CCI) rats could be prevented by pretreatment of ifenprodil. Methods: Male Sprague-Dawley rats weighing 180 - 200 g were divided into four groups, sham-operation + saline group; sham-operation + ifenprodil group; CCI + saline group and CCI + ifenprodil group. Peripheral mononeuropathy was produced by loosely ligating the rat's common sciatic nerve, a procedure which caused chronic constrictive injury of the ligated nerve. In sham-operated animals, the same dissection was performed but without ligation. Rats received 3 daily intraperitoneal (i. p. ) ifenprodil ( 10 mg/kg) or saline injections 30 min prior to nerve ligation according to the groups. The mechanical pain threshold of the rats before and after the operation were determined with avon Frey hair test. Results : CCI + saline rats exhibited lower mechanical pain threshold on days 7 and 14 after nerve ligation (P 〈0.05). But the rats of the other groups exhibited just the same mechanical pain threshold as that before the operations (P 〉 0.05 ). The CCI rats receiving pretreatment of ifenprodil exhibited higher mechanical pain threshold than that receiving pretreatment of saline (P 〈 0.05). Conclusion. The blockade of NR2B subunits of NMDA receptors can prevent the pain behavior of CCI rats. NR2B activation might be critical for the induction and/or maintenance of neuropathic pain following constrictive nerve injury.
