辣椒子叶离体培养及植株再生体系建立 被引量:12

摘要 以10个辣椒品种的子叶为试材,探讨了影响辣椒子叶离体培养及再生的各种因素(子叶外植体的生理年龄、外植体的部位、基本培养基、激素、硝酸银及光照强度等),筛选出辣椒子叶高效芽分化培养基为MS+6-BA5.0 mg/L+IAA1.0 mg/L+AgNO34.0 mg/L,平均芽分化率达到90.5%;高效芽伸长培养基为MS+6-BA3.0 mg/L+IAA1.0 mg/L+GA32.0 mg/L+AgNO34.0 mg/L;生根培养基为1/2 MS+IAA 0.2 mg/L+NAA0.1 mg/L,建立了辣椒高效再生体系。 Ten varieties of pepper (Capsicum annuum L. ) were selected as experimental materials. The effects of seedling stage, genotypes, hormone, organic nutrient component and AgNO3 et al on the tissue culture of pepper cotyledon explants were investigated. As a result, the best media for in vitro regeneration of cotyledon explants of pepper were discovered as follows: the bud induction medium is MS+6-BA 5. 0 mg/L+IAA 1. 0 mg/L+AgNO3 4. 0 mg/L; the bud elongation medium: MS+6-BA 3. 0 mg/L+IAA 1. 0 mg/L+GAa2. 0 mg/L+AgNOa 4. 0 mg/L;and the rooting medium: 1/2 MS+IAA 0. 2 mg/L+NAA 0. 1 mg/L Thereby a highly efficient regeneration system for pepper in vitro is established.
