摘要
分离纯化锥虫类原生动物细胞核存在着许多困难,阻碍了人们对这类单细胞生物的细胞核在各方面作进一步研究。本文提出一个从破碎细胞开始的分离核的完整程序。采用起始密度为1.083的Percoll工作液作等密度梯度离心,获得了纯度较高的利什曼原虫(Leishmania gerbilli)细胞核。经测定,L.gerbilli细胞核在Percoll中的浮力密度为1.048—1.051。
A complete procedure of nuclei isolation and purification from Leishmania gerbilli was established. With this procedure highly purified nuclei of L. gcrbilli can be obtained. The isopycnic density of the nuclei is about 1.048-1.051 in percoll media.
