摘要
目的:制备opticin特异性抗体并探讨opticin在牛眼中的分布。方法:将合成的多肽链免疫兔以获得OPT-C抗体,运用该抗体从玻璃体提取物中检测opticin,用免疫组织化学方法检测opticin在牛眼中的分布。结果:牛眼玻璃体提取物Western blot在40-50 kU可看到2个清晰可辨的分离的条带。牛眼免疫组织化学发现opticin存在于牛眼视网膜、玻璃体和睫状体无色素上皮中。结论:Opticin存在于睫状体中,支持opticin可能由睫状体无色素上皮细胞分泌这一推测。Opticin与玻璃体胶原纤维相连,可能与玻璃体结构的稳定有关。
Objective: To obtain specific antibody of opticin and to investigate the distribution of opticin in bovine eye. Methods: The distribution of opticin was detected from bovine vitreous extraction and paraffin embedded sections from bovine eyes were subjected to immunohistochemical analysis. Results: Western blot analysis of bovine vitreous specifically identified opticin as a discernible doublet at 40-50 kU. Opticin was immunolocalised to the retina, vitreous and ciliary body nonpigmented epithelium. Conclusion: Studies had previously demonstrated opticin expression by the non-pigmented ciliary epithelium. The immmunolocalisation data supported the proposition that the non-pigmented ciliary epithelium secrets opticin. The staining along the vitreous collagen suggests a connection between opticin and collagen, and opticin may be related to the stability of vitreous.
