丙型肝炎病毒NS4B对肝细胞增殖和细胞周期的影响 被引量:3

摘要 目的研究丙型肝炎病毒非结构蛋白4B(HCV-NS4B)对肝细胞细胞增殖和细胞周期的影响,探讨HCV-NS4B在HCV致病中的可能机制。方法利用脂质体介导将空白载体PCXN2及重组质粒PCXN2-NS4B转染入LO2肝细胞,G418筛选。RT-PCR鉴定质粒成功转染入肝细胞内。MTT法绘制生长曲线,观察NS4B对肝细胞生长的影响;流式细胞仪检测细胞周期的情况;免疫组化方法观察PCNA,cyclinD1的表达情况。结果NS4B转染入LO2有表达;绘制生长曲线示NS4B可促进肝细胞生长;细胞周期检测显示转染空载体PCXN2的细胞(65.28±6.23)%进入G0/G1期、(27.39±4.89)%进入S期、(7.34±4.54)%进入G2/M期;转染PCXN2-NS4B的细胞(46.89±5.60)%进入G0/G1期、(36.31±4.36)%进入S期、(16.80±6.79)%进入G2/M期;转染NS4B的细胞PCNA,cyclinD1表达率较转染空白载体组高,两组比较差异有显著性(P<0.01)。结论HCV-NS4B可能通过调节某些基因转录与表达,促进DNA合成,干扰肝细胞周期,促进肝细胞增殖。
