摘要 目的研究黄芪多部位组合对化学性及免疫性肝损伤小鼠的保护作用。方法采用D-氨基半乳糖胺(D-GalN)、卡介苗(BCG)+脂多糖(LPS)分别诱导小鼠化学性和免疫性肝损伤模型。L929细胞杀伤法检测肿瘤坏死因子(TNF-α)活性;TBA比色法测肝匀浆丙二醛(MDA)含量;[3H]TdR掺入法测脾淋巴细胞增殖反应及白细胞介素1(IL-1)的活性;赖氏法检测血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)的水平。结果黄芪多部位组合(60、120、240mg/kg)明显降低D-GalN小鼠升高的肝脏指数、血清ALT水平及肝匀浆MDA的含量;恢复刀豆蛋白A(ConA)诱导的免疫性肝损伤小鼠脾淋巴细胞增殖反应,抑制其腹腔巨噬细胞TNF-α、IL-1的产生;同时还能降低其血清ALT和AST水平。结论黄芪多部位组合对化学性及免疫性肝损伤小鼠均有明显的保护作用。 Objective To investigate the protective effects of Compound Astragalus Extract (CAE) on fiver injury in mice. Methods The acute fiver injury models were induced by D-galactosamine (D-GalN, 800 mg/kg, ip) and Bacillus Calmette Guerin (BCG,1× 10^7/0.2 mL, iv) plus Lipopolysaccharide (LPS 7.5 μg/0.2 mL, iv) in mice. At,T, AST, MDA content in fiver homogenate were assayed by sepctrophotometry. IL-1 activity and ConA-induced splenocytes proliferation were detected by PHI TdR incorporation assay. TNF- α activity was determined by assay of cytotoxicity against L929 cell. Results CAE (60, 120, 240 mg/kg) could obviously lower the elevated fiver index, ALT level in serum and MDA content in liver homogenate in D-GalN mice and in the immunological fiver injury mice. It also decreased ALT, AST level in serum, recovered ConA-induced splenocytes proliferation. It had inhibitory effect on inflammatory factors TNF, IL-1 secreted by PMΦs. Conclusion CAE showed significant protective effects on chemical and immunological fiver injury in mice.
