摘要
本文借助扫描电镜(Scanning Electron Microscope,SEM)对在家蚕胚胎细胞(Bombyx mori-SWU1Embryoniccell line,BmE-SWU1)中增殖的家蚕微孢子虫(Nosema bombycis)进行了观察;同时,利用间接免疫荧光试验(Indirect Immunofluorescence Test,IFAT)对家蚕微孢子虫极丝弹出情况进行了研究。结果发现:在家蚕微孢子虫体外培养体系中观察到了二型孢子、发芽后的孢子空壳以及未成熟孢子等;此外,在接种后10d的细胞爬片中,发现了大量孢子弹出极丝的情况,其极丝的长度在58μm-120μm之间,其中100μm以上的居多,且其极丝的扭曲程度、方向性、粗细也大都不同。本研究表明了微孢子虫在培养细胞体系中自动发芽可以以长极丝孢子的形式来完成。
This paper presents documentation of Nosema bombycis in an infected BmE-SWU1 cell line. The propagation of N. bombycis in infected BmE-SWU1 cells was observed by scanning electron microscopy. An indirect fluorescent-antibody assay was employed to investigate the discharged polar tubes of N. bombycis. Dichotocarpism of spores, empty spores and young spores were observed. A number of spores that had recently, or were still in process of extruding their polar tubes, were displayed. The discharged polar tubes ranged from 58 to 120 μm in length, and most were longer than 100μm. In addition, the degree of distortion, directivity and thickness of the polar tubes were extremely variable. Our results indicate that the spores of N. bombycis with long polar tubes ( 〉 100 ~m) can germinate automatically in cell culture systems [ Acta Zoologica Sinica 53 (6): 1107 - 1112, 2007].
