摘要 目的:探讨感毒清口崩片对流感病毒FM1致小鼠病毒性肺炎的细胞免疫作用机理。方法:以小鼠经鼻吸入流感病毒亚洲甲型鼠肺适应株(FM1)复制小鼠病毒性肺炎模型,用感毒清高、中、低剂量灌胃治疗5天,并设利巴韦林、莲花清瘟阳性对照及正常对照。采用SABC免疫组织化学法检测流感病毒感染小鼠肺组织细胞因子ICAM-1、NF-κB P65的表达。结果:感毒清高、中、低剂量组和利巴韦林、莲花清瘟组与模型组比较ICAM-1阳性表达明显减少(P<0.01);模型组NF-κB P65显著高于正常组(P<0.01)。各给药组NF-κB P65的活化程度(棕黄色颗粒)明显少于模型组(P<0.01)。结论:感毒清能抑制ICAM-1和NF-κB P65,从而抑制过度的炎症反应,减轻肺损伤。 Objective: To study the effects of Ganduqing buccal tablets (GDQ) on the mice with viral pneumonia infected by influenza virus FM1, and to explore the mechanism of cytoimmunity. Methods: Mice were inoculated intranasally with mouse lung-adapted influenza virus strain (FM1 ), and were treated with GDQ in high dose, middle dose and low dose for five days. Ribavifin and Lianhua Qingwen ( a kind of TCM decoction ) were used as positive control and blank drugs respectively. SABC immunohistochemical method was used to detect the expression of cytokine ICAM-1, NF-κB P65 in paraffin section of lung tissue of mice infected by influenza virus FM1. Results: The expression of ICAM-1 in GDQ group (including high-dose group, middle-dose group and low-dose group) was significantly lower than that in Ribavifin group, Lianhua Qingwen group and model group ( P 〈 0.001 ) . The expression of NF-κB P65 in model group was significantly higher than that in the blank group ( P 〈 0.01 ) . The activation level ( dark yellow beads) of NF-κB P65 in GDQ groups, Ribavirin group and Lianhua Qingwen group was lower than that in the model group ( P 〈 0.01 ) . Conclusion: GDQ can restrain the expression of ICAM-1 and NF-κB P65, restrain the excessive inflammatory reaction and finally lessen the lung injury.
