摘要
目的探讨端粒酶RNA反义寡核苷酸对舌癌细胞系Tca8113细胞增殖和端粒酶活性的影响。方法反义寡核苷酸(AS-ONS)作用于Tca8113细胞;采用MTT法测定AS-ONS对Tca8113细胞的毒性;倒置相差显微镜观察细胞生长状况及形态学改变;透射电镜观察细胞超微结构;测定Tca8113细胞端粒酶活性。结果AS-ONS可明显地抑制Tca8113细胞的生长,其生长抑制随剂量的增加而增强,具有浓度依赖性;AS-ONS对于Tca8113细胞端粒酶活性抑制作用具有时间依赖性。不同浓度AS-ONS作用于Tca8113细胞,其细胞毒性作用较弱;在倒置显微镜下可见AS-ONS组随药物浓度增高,细胞生长出现抑制。透射电镜观察到AS-ONS组作用的细胞细胞膜破损,细胞核内染色质凝聚,边集,并穿过核膜溢入胞质中,胞质部分溶解,线粒体空胞变性;AS-ONS对端粒酶活性抑制效果非常明显。结论反义寡核苷酸AS-ONS可明显地抑制Tca8113细胞的生长,并具有剂量依赖性和时间依赖性;靶向于端粒酶RNA的反义寡核苷酸可明显地抑制Tca8113细胞端粒酶活性,细胞增殖受到明显抑制。
Objective To investigate the effects of Telomerase RNA-targeting antisense oligonucleotides on Tca8113 cell proliferation and the cell telomerase activity.Melhods The toxicity of AS-ONS to Tca8113 cell was done by MTT method. The AS-ONS concentrations were 1.25,2.5,5.0,10.0,20.0/unol/L, respectively. The medium without serum and same volunm served as control. The OD value was measured at the wave length 570 nm.The Tca8113 cell treated with AS-ONS was observed under up-side-down microscope and electronic microscope.The Tca8113 cell telomerase activity was measured. Results Anfisense oligonucleotides could apparently inhibit the Tca8113 cell growth ( P 〈 0.01 ). Its effects were dose-dependent. Anfisense oligonucleotides effects were duration-dependent. The various AS-ONS concentrations acted on Tca8113 cell.The toxicity of AS-ONS was low.The Tca8113 cell treated with AS-ONS was growth-inhibited, observed under up-side-down mi- crescope and electronic micrescope.Antisense oligonucleotides could apparently inhibit the telomerase activity ( P 〈 0.01 ) and had duration- dependent effects. Conchmon Antisense oligonucleotides can apparently inhibit the Tca8113 cell growth. The inhibition effects are dose-de- pendent and duration-dependent. Telomerase RNA-targefing antisense oligonucleotides can significantly inhibit the Tca8113 cell telomerase activity and Tca8113 cell proliferation.
