摘要
目的探讨表皮生长因子(EGF)对大鼠先天性膈疝(CDH)模型肺发育活性物质的影响,了解EGF的可能作用机制。方法建立大鼠CDH模型。随机分为正常、CDH、CDH加EGF组,采用免疫组织化学方法检测各组胎肺组织甲状腺转录因子-1(TTF-1)、转化生长因子(TGFβ1)表达情况。结果TTF-1和TGFβ1在各实验组中均有表达,且二者在正常、CDH和CDH加EGF组的两两比较均有显著性差异(Pa<0.05)。随EGF剂量增加,TTF-1表达增强,而TGFβ1表达减弱,各组间均有显著性差异(Pa<0.05)。结论EGF对CDH大鼠胎肺组织TTF-1和TGFβ1表达部位和强度均有影响,可能是其影响CDH肺发育机制之一。
Objective To explore the effect of epidermal growth factor(EGF) on expressions of thyroid transcription factor - 1 ( TTF - 1 ) and transformal growth factor β1 (TGFβ1) in pulmonary hypoplasia in experimental congenital diaphragmatic hernia(CDH) model and learn the possible mechanism of EGF. Methods CDH was induced in rats by nitrofen. The experiment animals were randomly assigned into the following groups : control, CDH and EGF plus CDH group. Immunohistochemistry with TTF - 1 and TGFβ1 antibodies were used to determin the changes of TTF - 1 and TGFβ1 expressions in each group. Results The expressions of TTF - 1 and TGFβ1 could be detected in each subgroups, and statistically significant differe-nces existed for the expression of TTF - 1 and TGFβ1 between any two groups ( Pa 〈 0.05 ). With the increase of EGF dosage, the expression of TTF - 1 increased correspondingly, but TGFβ1 decreased, there were significant difference in each group ( Pa 〈 0.05 ). Condusion EGF can affect the expression locations and extensities of TTF - 1 and TGFβ1 in the CDH lung,which might be one of possible mechanisms affecting the pulmonary development.
