摘要
目的通过筛选一株高产β-葡萄糖苷酶菌株对栀子进行发酵,使得其中的京尼平苷转化为京尼平。方法通过底物特异显色筛选模型筛选菌株,并使用正交实验对发酵条件进行优化。结果筛选到一株产β-葡萄糖苷酶酶活可达到8.2 u/ml的青霉Penicillium nigricans,并优化了发酵转化条件,使得京尼平苷的转化率可达到95%以上。利用波谱法对发酵产物进行了结构鉴定,证明转化产物为京尼平。结论通过微生物转化的方法可以使栀子中主要成分京尼平苷转化成具直接作用的有效成分京尼平。
Objective Gardenia (Gardenia jasminoides Ellis ) was subjected to fermentation process by β-glueosidase strain to transform geniposide into genipin, a more effective compound. Methods The model of specific colorizing was used to screen the strain. The conditions of fermentation process were optimized by an orthogonal layout. Results Through the model of specific colorizing, the strain with high ability of microbial transformation has been screened. The transformation rate could reach to 95%. It was proved that the products was genipin. Conclusion Geniposide which was the effective compound of gardenia can be transformed into genipin by the method of microbial transformation.
