摘要
目的:研究蟾蜍灵诱导结肠癌细胞的凋亡作用。方法:采用不同浓度的蟾蜍灵处理结肠癌SW620细胞,采用四唑蓝比色法检测细胞生长,分别通过检测caspase-3活性和琼脂糖凝胶电泳了解细胞凋亡情况,以Fura-2荧光复合技术测结肠癌细胞内游离Ca2+浓度。结果:蟾蜍灵对结肠癌细胞增殖具有很强的抑制作用。蟾蜍灵能够诱导结肠癌细胞凋亡。在结肠癌细胞凋亡过程中,胞内游离Ca2+浓度明显升高,以凋亡早期更为明显。1 h时为(288.56±8.46)nmol/L,明显高于对照组(125.56±3.18)nmol/L(P<0.01)。结论:蟾蜍灵对结肠癌细胞生长具有很强的抑制作用,与诱导细胞凋亡有关。Ca2+在蟾蜍灵诱导结肠癌细胞凋亡过程中有一定的作用。
Objective: To study the effect of bufalin on colon cancer cell and explore its mechanism. Methods: After colon cancer cell line SW620 was treated with different concentration of bufalin, MTT was performed to evaluate the growth of cancer cell, and the apoptosis were determined with caspase-3 activity and agarose gel electrophoresis, respectively, and [Ca^2+] i was exmined by Fura-2 fluorescin load assay. Results: Bufalin had a remarkable inhibiting effect on colon carcinoma cell line SW620. Anticancer effect of bufalin on SW620 might be correlated with apoptosis. There exhibited apoptosis manner in cancer cells treated with bufalin,i, e, caspase-3 activity increased and DNA laddered. [Ca^2+] i was significantly in- creased, especially at the initial stage of apoptosis. It was(288.56±8.46 )nmol/L at 1 h,which was significantly higher than the control group(125.56±3.18 ) nmol/L(P 〈0.01 ). Conclusion: Bufalin can inhibit human colon carcinoma cell line SW620. The induction of apoptosis would be a very important mechanism in the treatment of colon carcinoma. [Ca^2+] i play an important role in the process of apoptosis by bufalin of colon carcinoma cell.
