2型糖尿病患者血清脂联素水平及脂联素基因SNP45分析 被引量:3

摘要 目的:探讨2型糖尿病患者的血清脂联素水平及脂联素基因第45位点单核苷酸多态性(SNP45)分布状况。方法:采用放免法对180例2型糖尿病组(T2DM组)患者和144例对照组(Con组)人群进行血清脂联素水平测定,并用TaqMan探针技术进行脂联素基因SNP45分析。结果:T2DM组血清脂联素水平明显低于Con组(P<0.05),T2DM组血清脂联素水平与各变量间的相关分析显示空腹血清脂联素水平与三酰甘油、空腹胰岛素原呈负相关,与HDL-C呈正相关;多元逐步回归分析显示:HDL-C是影响T2DM患者血清脂联素水平的独立相关因素。脂联素基因多态性分析:SNP45在T2DM组与Con组比较差异有统计学意义(P<0.05),等位基因分布频率提示G等位基因在糖尿患者人群中多见(P<0.05),糖尿病组中携带TG+GG型的患者血清脂联素水平显著低于TT型的患者(P<0.05)。结论:糖尿患者群中G等位基因多见,携带等位基因G的患者血清脂联素水平降低,提示脂联素基因SNP45多态性与2型糖尿病发病有关。 Objective: To investigate the state of serum adiponectin level and adiponectin gene SNP45 in type 2 diabetes patients. Methods : Serum adiponectin levels of 180 patients with type 2 diabetes ( T2DM group) and 144 participants( Con group) were assayed by RIA, and adiponectin gene SNP45 of them were analysised by TaqMan technology. Results: The levels of serum adiponectin in T2DM group were signifi- cantly lower than those in Con group(P 〈 0.05 ). Correlation analysis between serum adiponetin level and other variances revealed that it was negatively associated with triglyceride, fasting serum proinsulin level, and positively associated with HDL-C. Stepwise multiple regression analysis revealed that HDL-C was inde- pendently correlated with the serum adiponectin level in T2DM group. The distributions of SNP45 genotypes were different between T2DM and Con groups. The analysis of allele frequency showed the major allele was G in diabetic patients. The subjects with SNP45 TG or GG genotype had lower serum adiponectin levels than did those with TI' genotype in type 2 diabetes. Conclusion: The major allele was G in type 2 diabetic patients. Serum adiponectin levels were lower in the subjects with allele G suggested SNP45 in adiponectin gene was associated type 2 diabetes.
