摘要
目的建立ig桂枝茯苓胶囊后Beagle犬血浆中丹皮酚的测定方法,并研究其药动学。方法给予Beagle犬桂枝茯苓胶囊后,于不同时间前肢静脉取血,制备血浆;血浆样品经小柱固相萃取,采用HPLC法测定血药浓度,统计矩分析方法计算药动学参数。结果丹皮酚的线性范围为25.0~1.6×10^3 ng·ml^-1,定量下限为25ng·ml^-1,日内RSD分别为9.2%、7.8%、2.5%;日间RSD分别为5.7%、2.4%、1.4%;Beagle犬给予桂枝茯苓胶囊后,血浆中丹皮酚的药-时曲线存在双峰,tmax1和tmax2分别为25、240min,Cmax1和Cmax2分别为195.5±90.3、69.8±17.1ng·ml^-1,AUC=414.4±35.8h·ng·ml^-1,MRT=3.2±0.4h。结论所用方法符合生物样品的测定要求,可用于Beagle犬体内丹皮酚血药浓度的测定和药动学研究。
OBJECTIVE To develop a method with diode array detector for determination of paeonol in Beagle dog plasma after ig Guizhi Fuling capsule(GFC) and study its pharmacokinetics. METHODS Beagle dogs were given GFC by ig. The plasma were collected at designed times from forelimb vein. Following solid - phase extraction, the blood concentration was determined by HPLC and its pharmacokinetics parameter was calculated by statistic moment analysis method. RESULTS The linear range was 25.0 - 1.6×10^3 ng·ml^-1. The lower limit of quantification was 25 ng·ml^-1. The intra - day variations of RSD were 9. 2% ,7. 8% ,2. 5% ,respectively. The inter - day variations of RSD were 5.7% ,2. 4% , 1.4% respectively. The phenomenon of double peaks was presented in the blood concentration -time curves of paeonol after ig GFC ,tmax1 and tmax2 were 25 min and 240 min, Cmax1 and Cmax2 were 195.5 ± 90. 3 and 69. 8 ± 17. 1 ng·ml^-1, respectively. CONCLUSION The method is specific, accurate, and sensitive enough to be applied to the pharmacokinetic studies of paeonol.
