摘要
真核系统表达的猪α干扰素(PoIFN-α)发酵液,经离心、收集上清液、透析过滤处理,通过FPLC-HisTrap HP Kit亲和层析系统分离纯化,最后以SDS-PAGE银染法检验。结果表明,在不同pH和NaCl浓度情况下,发酵液经亲和层析系统分离纯化后均可以得到较为单一的目的蛋白组分,而在pH8.2,NaCl0mmol/L的情况下得到的目的蛋白更纯,且非特异性吸附也较少。
The recombinant porcine-α interferon (PolFN-α) expressed by eukaryotic cells was collected and purified by centrifugation, filtration, and FPLC-HisTrap HP Kit affinity chromatography system, and finally assayed by silver staining. It was shown that in the conditions of different pH val- ues and NaC1 concentrations a single band of target protein could be obtained after the fermented broth was separated and purified through affinity chromatography system, and more purified target protein could be obtained in the conditions of pH 8.2 and 0 mmol/L NaCl.
