摘要
生长分化因子5(Growth and differentiation factor 5,GDF-5)是人体软骨发生和骨骼发育过程中发挥重要功能作用的一种调节因子,其基因改变可引起诸多肢体畸形,如Hunter-Thompson肢端发育不良、A2或C型短指/趾症、Grebe型软骨发育不良、腓骨发育不全/复合性短指、多发性骨性联合综合征、近端指/趾关节粘连和骨关节炎,因此近年来受到了广泛关注。此外,其在组织损伤修复、神经系统保护等方面的广阔应用前景越来越被专家学者们所重视。本文将就GDF5在肢体骨骼发育中的作用、GDF5突变引起的相关疾病、GDF5在组织工程学上的应用现状等方面进行综述,并对GDF5未来的研究与应用前景进行了探讨。
Growth and differentiation factor 5 (GDF-5) plays key regulatory roles on both chondrogenesls and osteogenesis. Alteration of GDF-5 gene can elicit a group of extremity malformation, such as acromesomelic dysplasia (Hunter-Thompson type),brachydactyly(A2 or C type),chondrodysplasia(Grebe type),fibular hypoplasia/complex brachydactyly, multiple synostoses syndrome, proximal sympphalangism and Osteoarthritis. Moreover, researchers pay more attention to its broad application prospect on wound healing and protection of nerve system. This review focuses on the functions of GDF-5 in osteogenesis, dieases induced by GDF-5 gene mutation and application of GDF-5 in tissue engineering. In additon, the future prospect on research and application of GDF-5 is also involved.
