摘要 应用超离心和柱层析结合聚丙烯酰胺凝胶电泳(PAGE)及电镜技术,研究中国鲎血蓝蛋白的结构特点.结果表明,经SephadexG-100纯化的血蓝蛋白在PAGE电泳中出现4条带.纯化的血蓝蛋白再经DEAE-32纤维素柱层析得到5个洗脱峰,每个峰在PAGE电泳下可分辨出4条带.在电镜下,血蓝蛋白分子出现环形、五角形、十字形和蝴蝶结形等构型并与其他解离的中间的构型同时存在. By using ultracentrifugation,chromatography,polyacrylamide gel electron phoresis (PAGE) and electron microscopy,the structural characteristics of hemocyanin of T.tridentatus have been investigated.The results show that there are four bands in PAGE for hemocyanin purified by Sephadex G 100 chromatography. Purified hemocyanin can be separated by means of DEAE 32 cellulose chromatography into five peaks, marked as I~V.Four bands can be distinguished by PAGE for each peak.Under electron microscope,five typical view of T.tridentatus hemocyanin molecule can be observed according to different project angle.They look like ring,pentagon,cross,and bowtie in shape respectively.There are some of intermediate dissociating configuration of hemocyanin that can be seen in the same field.
