摘要
目的利用基因表达谱芯片技术,寻找人类肺鳞癌组织和正常肺组织中与生物信号转导或免疫反应有关的差异表达基因。方法分别提取肺鳞癌和正常肺组织总RNA后,逆转录合成cDNA探针,混合后与Biostar H-Ⅰ型基因芯片杂交,ScanArray 4000扫描仪扫描杂交信号,计算机分析差异表达基因。结果筛选出在3组以上标本中存在差异表达的基因77条,其中44条表达上调(r〉2.0),33条表达下调(r〈0.5)。结论利用基因芯片技术实现了高通量筛选与肺癌发生发展密切相关的基因,发现了77条在肺鳞癌中与细胞信号转导或免疫反应有关的差异表达基因,为今后更深入的研究指明了方向。
Objective By use of cDNA microarray,to screen out the differential expression genes which are related with biological signal conduction,humoral or cellular immunity between lung squamous cell carcinoma and normal lung tissue.Methods The total RNAs were isolated from tumor and normal lung tissue and were reversely transcribed to prepare the hybridization probe.The mixed probes were hybridized to the BiostarH-Ⅰ cDNA microarray.After washing,the cDNA microarray was scanned for the fluorescent signals by ScanArray 4000 scaning apparatus.The scanning data were analyzed by Genepxix Pro software and differential expression genes between the two kinds of tissues.Results Seventy-seven genes were screened out for differential expression genes in more than three groups of sampling.Among them,fourty-four genes showed up-regulated expression(r〉2.0),and the others down-regulated expression(r〈0.5).Conclusions Our studies showed that cDNA microarray can help us to realize high-throughput screening of the genes associated with lung cancer.Eight genes which are related with biological singal conduction or immunity in lung squamous cell carcinoma were found.Further investigation about these genes will be carried out later.
