摘要
目的研究灵芝孢子对帕金森病(PD)动物模型黑质、纹状体神经递质的影响,以探讨灵芝孢子对帕金森病的脑保护作用。方法120只SD大鼠随机分为3组,PD组:立体定向注入6-羟多巴,术后1周腹腔注入阿朴吗啡观察30min大鼠旋转的次数,连续至第4周每分钟大于6次者为成功的帕金森氏病模型;灵芝孢子组:先用灵芝孢子粉灌胃3d,立体定向注入6-羟多巴,继续灌胃4周,直至处死。正常对照组:立体定向注入黑质抗坏血酸生理盐水。处死后快速取术侧中脑、纹状体,匀浆,高效液相色谱仪检测中脑黑质、纹状体神经递质多巴胺(DA)、3,4-双羟苯乙酸(DOPAC)、高香草酸(HAV)、5羟色胺(5-TH)、去甲肾上腺素(NE)含量等的变化。结果高效液相色谱仪结果显示PD组黑质、纹状体的DA、DOPAC、HAV、5-TH、NE含量较正常对照组明显降低,灵芝组术侧黑质、纹状体DA、DOPAC、HAV、5-TH、NE含量较PD组明显增高。结论灵芝孢子能够逆转PD组DA、DOPAC、HAV、5-TH、NE含量的下降,对PD有脑保护作用。
Objective To examine the neuroprotective role of ganoderma spores in a Parkinson's disease (PD) rat model. Methods 120 healthy male Sprague-Dawley rats were equally divided into three groups randomly. Hemiparkinsonian model was established by unilateral stereotaxic infusion of 6-hydroxydopamine (6-OHDA) into the right substantia nigra of midbrain. After operation, rotational behavior test was performed weekly. The number of rotations over a period of 30 minutes was recorded after intraperitoneally injection with apomorphine. Rats showing a rotation of more than 6 circles per minute at the forth week after operation were considered PD rats. The second group of rats was given oral ganoderma spores for three days before the introduction of lesion. After the operation, ganoderma spores were given orally for another 4 weeks. The third group of rats was injected with natrium saline (control). At the end of the experiment, the rats were killed, and the middle brain was removed and homogenized. The expressions of dopamine(DA), 3,4-dihydroxyphenylacetic acid(DOPAC), 4-hydroxy-3-menthoxyphenylacetic acid( HVA ), 5-hydroxytryptamine ( 5-HT), and noradrenalin(NE) were analyzed by high performance liquid chromatography(HPLC). Results Comparing with the PD group, the levels of DA, DOPAC, HAV, 5-TH, and NE in substantia nigra and in the middle brain of the ganoderma spores treated rats were significantly increased. Conclusion Oral feeding of rats with ganoderma spores can increase the expression of DA, DOPAC, HAV, 5-TH, and NE. The results suggested that ganoderma spores may have a neuroprotective function in PD rats.
