压应力对增生性瘢痕成纤维细胞生长特性的影响 被引量:5

摘要 目的:探讨不同持续时间压应力对人增生性瘢痕成纤维细胞生长特性的影响。方法:组织块培养法获取人增生性瘢痕成纤维细胞(HSFb),并随机将HSFb随机分为对照组(未施压)、持续施压组(25mmHg,6~8天)和施压(25mmHg,3~4天)后去除压应力组。分别以MTT法测定细胞生长抑制率、流式细胞仪测定细胞生长周期、免疫组化观察增殖细胞核抗原(PCNA)的表达及Annexin-V-PI标记法测定细胞凋亡情况。结果:25mmHg持续压应力对HSFb的生长抑制率随加压时间的延长逐渐增大;与对照组相应时相点比较,PCNA表达降低,处于增殖期(S+G2+M)细胞比例减小;细胞凋亡率随施压时间延长逐渐增加;而施压后去除压应力继续培养观察显示,HSFb的抑制率逐渐回降,PCNA表达增加和增殖期细胞比"例反弹"增大,细胞的凋亡率也逐渐下降。结论:适当的持续压应力对人增生性瘢痕成纤维细胞具有抑制增殖与促进凋亡的共同效应,短期加压后去除压力致使上述效应"反弹性"降低,可能是临床上对增生性瘢痕压迫治疗需要早期、持续、长期维持的原因之一。 Objective To investigate the effects of mechanical stress on the growth characteristics of human hypertrophic scar fibroblasts (HSFb) in vitro, Methods Human hypertrophic scar fibroblasts obtained from the patient who suffered from hypertrophic scar in my hospital were isolated and cultured in vitro, and then they were randomly divided into 3 groups as follows=control group (no compression), continuous compression group ( with 25 mmHg air pressure for 6-8 days ) and the cancelled stress after compressing group. The cellular proliferation was determined by MTT assay,cell cycle was detected on the flow cytometer,and PCNA expression in HSFb was confirmed by immunohistochemistry method. The apoptosis of scar fibroblasts was measured with Annexin V binding with PI labeling. Results Both the inhibiting rate of HSFb growth and the apoptosis were markedly increase in continuous pressure group with loading time, Compared with control group,the PCNA expression and cell ratio within proliferative phase (S+ G2+M) was decreased. However,the inhibiting ratio of HSFb growth,PCNA expression,cell ratio within proliferative phase and apoptosis ratio had gradually returned in the group of cancelled load after pressure. Conclusion Based on the above results,we suggested that combined effects of the proliferation inhibition and apoptosis promotion induced by continuous pressure in HSFb and the return of that combined effects after cancelling stress may potentially be one of the important reasons of well obtained therapeutic effect with pressure therapy for needing early stage and long time maintenance during the clinical practice,
