摘要 目的探讨G蛋白与增生性瘢痕瘙痒发生的关系及可能的机制。方法Ⅰ型胶原酶和透明质酸酶消化分离肥大细胞,用荧光分光光度法检测G蛋白失活剂NEM对人增生性瘢痕、非增生性瘢痕以及皮肤中肥大细胞释放组胺的影响;RT-PCR法分析P物质(SP)对Gi-2αmRNA表达的影响。结果①与对照组比较,SP均明显增强三种组织中肥大细胞分泌组胺的效应,而NEM对其效应有明显的抑制作用。②对照组中,增生性瘢痕肥大细胞中Gi-2αmRNA的表达明显弱于非增生性瘢痕和正常皮肤(P<0.01);SP作用后,Gi-2αmRNA在三种组织中的表达与对照组比较差异无显著性。结论G蛋白介导了SP刺激人皮肤和瘢痕组织中肥大细胞分泌组胺的效应;Gi-2αmRNA的弱表达可能在增生性瘢痕瘙痒发生中起重要作用。 Objective To study the relationship between G protein and the itching of hypertrophic scar. Methods Mast cells(MCs) were separated by collagenaseI and hyaluronidase. Fluorospectrophotometry was used to examine histamine after inducing G protein inactivated agent N-ethyl maleimide(NEM) in the MCs of hypertrophic scar (HS), non-hypertrophic scar(NHS) and normal skin. Gi-2αmRNA was semi-quantitatively analyzed after RT- PCR. Results After MCs of HS, NHS and normal skin pretreating with NEM, the effect of SP on the by release of histamine was significantly inhibited( P 〈0.01 ). The expression of Gi-2αmRNA in MCs of HS was significant lower than those of NHS and normal skins in control group( P 〈0.01 ), while there was little difference of Gi-2αmRNA expression in HS, NHS and normal skin MCs between SP group and control group. Conclusion G protein partly mediate the histamine releasing effect of human scar and skin MCs stimulated by SP; The low expression of Gi- 2αmRNA in HS MCs maybe play important role in the itching.
