摘要
目的从吉日很阿嘎如-8中提取,精制多糖;并研究其抗氧化性能。方法吉日很阿嘎如-8经热水抽提,Sevag法脱蛋白,乙醇沉淀,再通过透析得白色粉末状精制多糖。并采用比色法测定其对·OH和O2-·的清除能力,并用甘露醇进行比较。羟基自由基(.OH)由Fenton体系产生,超氧阴离子自由基(O2-·)由邻苯三酚自氧化体系产生。结果吉日很阿嘎如-8多糖对.OH和O2-.具有一定的清除能力,其达到50%清除率所需药物浓度EC50分别为0.81,2.80mg/ml。同样条件下甘露醇达到50%清除率所需药物浓度EC50分别为2.65,2.64mg/ml。结论吉日很阿嘎如-8多糖能有效清除·OH和O2-·,其清除能力较甘露醇强,并且浓度与清除率呈正相关。
Objective To study the extraction, purification and antioxidant effect of polysaccharides from Jirihen Agaru - 8. Methods Jirihen Agaru -8 polysaccharide was extracted with water. The herb was deproteined, ethanol precipitated, and purified with dialyses method. Its antioxidant effect was tested by the colorimeter and compared with Nectar. Hydroxyl ( · OH) was produced by Fenton reaction and super oxide radical ( O2^-· ) was produced by the oxidation of pyrogallol. Results Polysaccharides of Jirihen Agaru - 8 had strong scavenging effect for ·OH and O2^-· The amounts of scavenging 50 % ( EC50 ) · OH and O2^-· was 0.81 mg/ml and 2.80 mg/ml. But the amount of scavenging 50 % (EC50) · OH and O2^-· was 2.65 mg/ml and 2.64 mg/ml of Nectar. Conclusion Jirihen Agaru - 8 polysaccharides has an action to scavenging · OH 和O2^-· Its activities is strong to Nectar, and presents positive correlationship with the amounts.
