姜黄素和环氧合酶-2抑制剂NS-398对肝星状细胞时效的双向调节体外实验研究 被引量:1

摘要 [目的]观察抗氧化剂姜黄素(Cur)与选择性环氧合酶-2(COX-2)抑制剂NS-398两种药物同时联用或分时联用对大鼠肝星状细胞(HSC)生长的影响。[方法]体外培养大鼠肝星状细胞株HSC-T6,接种96孔板,分为5组。Ⅰ:单独姜黄素系列浓度组(0、20、30、40μmol/L);单独NS-398系列浓度组(0,20,40,80μmol/L);Ⅱ:单独NS-398系列浓度组(ABSI)和NS- 398系列浓度组+姜黄素20μmol/L(ABS2)对大鼠HSC增殖的影响,Ⅲ:两种药物同时加药组;Ⅳ:两种药物先后6 h加药组:先加姜黄素20μmol/L作用HSC6h后加NS-398系列浓度组(0、20、40、80μmol/L);先加NS-398系列浓度(0、20、40、80μmol/L)作用HSC6h后加姜黄素20μmol/L组,Ⅴ:析因分析组。各组设不加药物的细胞对照组,与HSC共同培养,以MTT法检测姜黄素、NS-398单用或同时、分时联用对HSC生长的影响。LDH法检测各组对HSC的细胞毒性作用。[结果]单独NS-398系列浓度组和单独姜黄素系列浓度组有抑制HSC的作用,但姜黄素的作用比NS-398强:同时加两药组比分时加药组有明显不同,先加NS-398作用HSC6h组比先加姜黄素作用HSC6h组的抑制率明显降低。[结论]时间因素导致两药联用是双向调节,同时加药或先加姜黄素作用HSC6h组后加NS-398时呈正调节作用,抑制率升高,而先加NS-398作用HSC6h后加姜黄素呈负调节作用,抑制率降低。NS-398对HSC作用一定时间后有抑制姜黄素对HSC起生物效应的作用。
