雷公藤内酯醇对AngⅡ介导的大鼠GMC增殖的影响 被引量:1

摘要 目的:观察不同剂量雷公藤内酯醇对血管紧张素Ⅱ(AngⅡ)介导的肾小球系膜细胞增殖的影响,探讨其可能机制。方法:分离培养大鼠GMC,用10-6mol/LAngⅡ刺激GMC,将培养的大鼠GMC随机分为5组:正常培养对照组、AngⅡ刺激组、AngⅡ和3种不同浓度(15、10、5μg/L)的雷公藤内酯醇共孵育组。分别采用MIT法及流式细胞术检测大鼠GMC增殖及细胞周期的变化。结果:雷公藤内酯醇能抑制AngⅡ介导的大鼠GMC增殖,降低S+G2/M期细胞比例,其中以高浓度雷公藤内酯醇作用最强(P<0.01)。结论:雷公藤内酯醇可能通过减少DNA合成期(S期)细胞数量,抑制大鼠GMC增殖。 Objective:To observe the effect of triptolide on the proliferation of the rat's mesangial cells induced by angiotensin,to discuss the possible mechanism of triptolide's inhibiting effect on the proliferation of the GMC.Methods:Extracting the GMC from healthy SD rat,culturing and passing cells.Cells were divided into five groups:control,AngII,and triptolide three groups.The proliferation of rats' GMCwas observed by MTT,the cell cycle change of the GMC was determined by flow cytometer.Results:Triptolide could inhibit the proliferation of rats' GMS induced by AngII,down-regulate the ratio of the S+G2/M phase of GMC.Conclusion:Triptolide might be able to inhibit the GMC's proliferation by decreasing the ratio of the DNA synthesis period in the GMC.
