生长阻滞特异基因1在胃癌细胞系和组织中的差异表达及其意义

摘要 目的探讨Gas1在胃癌细胞系和组织中的差异表达及与胃癌发生、发展的关系。方法应用RT-PCR检测Gas1在7个胃癌细胞系和23对胃癌配对组织标本中的表达情况,免疫组化检测Gasl在43例手术切除的胃癌标本和20例正常胃黏膜组织中的表达情况。结果7个胃癌细胞系均不表达Gas1mRNA;23对胃癌配对组织标本中癌旁组织Gas1mRNA阳性表达率高于胃癌组织(P<0.01);20例正常胃黏膜组织中有19例表达Gasl蛋白(阳性表达率为95.0%),43例胃癌标本中有2例表达Gasl蛋白(阳性表达率为4.65%),两者相比差异具有显著性(P<0.01)。结论Gasl在胃癌细胞系中表达缺失,可能与胃癌细胞系的细胞学特征有关;Gasl在胃癌表达缺失,可能与胃癌的发生发展有关,Gasl可作为判断胃癌生物学行为的一个有用指标。 [Objective] To study the expression of Gasl and its significance in gastric carcinoma cell hnes and mucosa. [Methods] RT-PCR was used to detect the expression of Gasl mRNA in gastric carcinoma cell lines and 23 paired tissues of gastric carcinoma mucosa, and immunohistochemistry was used to detect the expression of Gasl protein in 20 normal gastric mucosa and 43 gastric carcinoma. [Results] The positive rate of Gasl mRNA in gastric carcinoma cell lines was 0; and 78.26% in poricarcinoma mucosa, 8.70% in gastric carcinom, respectively. Gasl mRNA of gastric carcinoma was significantly lower than that of pericarcinoma (P 〈0.01). The positive rate of Gasl protein in normal mucosa was 90.0 % compared with 4.65 % in gastric carcinoma. [Conclusion] The expression of Gasl might be related to the carcinogenesis, development, and prognosis in gastric carcinoma.
