摘要
目的:探讨硝苯地平对人牙龈成纤维细胞Ⅰ型胶原合成的影响。方法:用含浓度为108、360、1200μg/L硝苯地平的DMEM液作用体外原代培养人牙龈成纤维细胞;酶联免疫吸附试验检测细胞Ⅰ型胶原表达水平;细胞计数观察细胞增殖。结果:Ⅰ型胶原合成检测和细胞增殖试验的结果都显示在第5天,高浓度1200μg/L、360μg/L组与低浓度108μg/L之间存在显著性差异。结论:高浓度硝苯地平有促进人牙龈成纤维细胞Ⅰ型胶原合成和细胞增殖的作用。
Objective:To investigate the effect of nifedipine,a calcium channel blocker,on the expression of collagen in gingival fibroblasts in vitro. Methods:Primarily gingival fibroblasts were cultured and incubated with various concentrations of nifedipine (108、360、1 200 μg/L). Enzyme-linked immunosorbent assay was used to evaluate the amount of type I collagen. Cell proliferation was measured by cell counting. Results:The expressions of collagen and cell proliferation were significantly different among the high concentration groups and the others on 5 th day,especially higher in 360 μg/L and 1 200 μg/L group. Conclusion: Nifedipine can increase the expression of type Ⅰ collagen in gingival fibroblasts and cell proliferation.
