摘要
目的用^(99m)Tc-亚甲基二膦酸盐(^(99m)Tc-MDP)联合胶体磷酸铬^(32)P治疗大鼠佐剂型关节炎,探讨其疗效并与单一使用^(99m)Tc-MDP或胶体磷酸铬^(32)P治疗进行比较。方法利用佐剂诱导SD大鼠的关节炎模型(AA),给予胶体磷酸铬^(32)P踝关节腔内注射以及^(99m)Tc-MDP腹腔注射,在不同的时间点观察大鼠左踝关节的左右径宽度、血清肿瘤坏死因子(TNF)和白细胞介素-1β(IL-1β)水平及关节病理的变化。结果联合治疗组大鼠左踝关节左右径宽度较^(32)P胶体治疗组小[第4周(7.11±0.34)mmvs(7.57±0.29)mm,P<0.01]。联合治疗较^(32)P胶体治疗更能有效地降低血清TNF[第4周时(1.6±0.4)ng/mlvs(2.0±0.3)ng/M,P<0.05]和IL-1β[第4周时(0.271±0.033)ng/mlvs(0.308±0.024)ng/ml,P<0.05;第6周时(0.209±0.023)ng/mlvs(0.255±0.016)ng/ml,P<0.01]的水平。在病理检查上,联合治疗组滑膜增生程度较^(99m)Tc-MDP治疗组轻,而炎细胞浸润程度较^(32)P胶体治疗组轻。结论^(99)Tc-MDP联合胶体磷酸铬^(32)P治疗大鼠佐剂型关节炎较单用^(99m)Tc-MDP或胶体磷酸铬^(32)P治疗效果更好。
Objective To compare the therapeutic effect of the combination of ^99mTc—methnetium methylene diphosphonate and colloidal chromic phosphate ^32p with the monotherapies in rats with adjuvant arthritis (AA). Methods In this study, 50 Sprague-Dawley rats were randomly divided into five groups with 10 rats in each: normal control group (arthritis induced and treated with the same volume of normal saline via the same administration route), AA control group (arthritis induced with adjuvant and treated with normal saline), ^32p colloid group (arthritis induced with adjuvant and treated with single intra-articular injection of colloidal chromic phosphate ^32p 0.74 MBq and intra-peritoneal injection of normal saline every other day), ^99mTc— MDP group (arthritis induced with adjuvant and treated with intra-peritoneal injection of ^99mTc—MDP 2.5×10^3μg/kg every other day and single intra-articular injection of normal saline), combination group (arthritis induced with adjuvant and treated with the combination of the two drugs). The diameter of the left hind ankle, serum levels of tumour necrosis factor (TNF) and interleukin-1β (IL-1β), and the pathological changes of ankle joints were tested at different time points. Results The diameter of the left hind ankle at week 4 was smaller in the combination group than that of the ^32p colloid treatment alone group [ (7.11±0.34)mm vs (7.57±0.29)mm, P〈0.01]. The combination was more efficacious than 32p colloid alone in decreasing the serum levels of TNF [(1.6±0.4) ng/ml vs (2.0±0.3) ng/ml, P〈0.05 for week 4] and IL-1β [(0.271±0.033)ng/ml vs (0.308±0.024)ng/ml, P〈0.05 for week 4]; [(0.209±0.023)ng/ml vs (0.255±0.016)ng/ml, P〈0.01 for week 6]. In histological findings, the combination therapy showed less synovium proliferation when compared with ^99mTc— MDP alone and fewer inflammatory cells infiltration when compared with ^32p colloid alone.
