摘要
应用氯化铵裂解、超声波破碎和离子交换层析等方法,从鹿白细胞中提取出阳离子抗菌肽混合物。经平板抑菌试验初步检测其抗菌活性后,采用微量反应板法测定了鹿抗菌肽对大肠杆菌、金黄色葡萄球菌、白色念珠菌等9个标准菌株的最小抑菌浓度(MIC),结果显示,该混合抗菌肽对革兰氏阳性细菌、革兰氏阴性细菌和真菌均具有抑制活性。体外杀菌曲线试验表明,鹿抗菌肽的杀菌作用是非浓度依赖性的,杀菌效果迅速,可在2 min内杀灭63.8%的受试细菌。
Cationic antimicrobial peptides were extracted from deer neutrophils through NH4Cl lyses, sonication and ion-exchange chromatography. Minimal Inhibition Concentrations (MICs) of the extract were determined against 9 standard strains such as E. coli, S. aureus and C. albicans by means of microdilution. The results showed that the extract of peptides was active for Gram-positive, curve experiments revealed that the killing potential of the Gram-negative bacteria and yeast. The in vitro time-kill peptides was concentration-independent and immediate, with 63.8% of the test bacteria killed in 2 minutes.
