实验性矽肺大鼠外周血和脾T淋巴细胞亚群的动态变化 被引量:11

Changes and Significance of T Lymphocyte Subsets in Peripheral Blood and Spleen at Different Stages in Silicotic Rat Model

摘要 [目的]研究实验性矽肺大鼠在染尘后不同时间点外周血和脾组织T淋巴细胞亚群数量的变化,并探讨其意义。[方法]将60只健康雄性SD大鼠随机分为染矽尘组和对照组,每组30只。对染矽尘组以非暴露方式气管内分2次(间隔1周)分别注入1ml二氧化硅悬液(20 mg/ml)建立大鼠矽肺模型,对照组气管内注入等量的灭菌生理盐水。染尘后第7、14、21、28、35、42、49、56、63和70天各处死实验组和对照组大鼠3只,采集外周血和脾脏,用流式细胞术检测大鼠外周血、脾组织的T淋巴细胞(CD3、CD4、CD8细胞)百分率。[结果]矽肺大鼠外周血CD4水平在实验早期(第1~3周)高于对照组。在实验后期,CD3、CD4、CD4/CD8明显低于对照组(P<0.05),CD8明显高于对照组(P<0.05)。脾组织CD3水平在第1~3周明显高于对照组(P<0.05),CD8水平在第1~3周轻度增高(差异无显著性)。在实验后期,CD3、CD4、CD4/CD8明显低于对照组(P<0.05),CD8明显高于对照组(P<0.05)。[结论]在实验早期,石英增强获得性免疫反应;在实验后期,矽肺大鼠外周血及脾T淋巴细胞亚群存在明显异常,细胞免疫功能严重紊乱,石英降低了获得性免疫反应。 [ Objective ] To investigate changes and their significance of T lymphocyte subsets in peripheral blood and spleen tissue at different stages after silica exposure in rat. [ Methods ] Sixty healthy male SD rats were randomly divided into the silica exposure group and the control group. 1 ml silica suspension ( 20 mg/ml ) was administrated via intratracheal instillation twice at an interval of one week while equal amount of saline was instilled to the rats of control group. Animals were sacrificed at the 7% 14^th 21^th 28th, 35^th, 42^th, 49^th 56^th 63^rd, and 70^th day after last instillation, and then the samples of peripheral blood and spleen were collected. The levels of T lymphocyte subsets in peripheral blood and spleen were detected by flow cytometry. [ Results ] The levels of CD4 in peripheral blood of silicotic rats were higher than those in control group in early stage( 1- 3 w ). The levels of CD3, CD4 and the ratio of CD4/CD8 obviously decreased ( P 〈 0.05 ), and the levels of CD8 significantly increased ( P 〈 0.05 ) in later stage of experiment. The levels of CD3 in spleen of sificotic rats significantly increased than those in control group in 1 - 3 w( P 〈 0.05 ), and the levels of CD8 increased slightly in 1 - 3 w( P 〉 0.05 ). In later stage of the experiment the levels of CD3, CD4 and the ratio of CD4/CD8 were obviously lower( P 〈 0.05 ), and the levels of CD8 were significantly higher than those in control group( P 〈 0.05 ). [ Conclusion] In the early stage of silicosis, silica increased adaptive immune responses. In the later stage, the levels of T lymphocyte subsets in peripheral blood and spleen of of silicotic rats were obviously abnormal. The function of cell immunity was in severe disorder. Silica decreased adaptive immunnity.
