摘要
目的:研究人黄素化颗粒细胞中肝素结合表皮生长因子(HB-EGF)的表达特点。方法:收集体外受精-胚胎移植过程中的人黄素化颗粒细胞进行体外培养;应用RT-PCR法和免疫细胞化学法分别检测HB-EGF的mRNA和蛋白表达;应用实时RT-PCR法检测HB-EGFmRNA量的变化。结果:人黄素化颗粒细胞中存在HB-EGF的mRNA和蛋白表达。培养基中加入重组HB-EGF后,HB-EGFmRNA的表达量迅速上升,2h内达高峰,然后回落,于12h内降至处理前水平。结论:人黄素化颗粒细胞中存在HB-EGF的表达;人黄素化颗粒细胞中的HB-EGF可能是瞬时早期反应基因。
Objective: To investigate the expression pattern of heparin-binding epidermal growth factor (HB-EGF) in human luteinized granulosa cells. Methods: Human luteinized granulosa cells were collected from patients tmdergoing in-vitro fertilization-embryo transfer (IVF-ET). The luteinized granulosa cells were cultured and treated with human recombinant HB-EGF. The expressions of HB-EGF mRNA and protein were detected by reverse transcription polymerase chain reaction (RT-PCR) and immunocytochemical method, respectively. The changes in HB-EGF mRNA in response to recombinant HB-EGF stimulation were measmed by real time RT-PCR. Results: The expressions of HB-EGF mRNA and protein existed in luteinized granulosa cells. The expression of HB-EGF mRNA increased after the recombinant HB- EGF was added into the culture, medium,reached the peak after 2 hours,and then returned to baseline within 12 hours. Conclusion: HB- EGF is expressed in human luteinized granulosa cells as an immediate-early gene.
