摘要
目的探讨内毒素脂多糖(1ipopolysaccharide LPS)在体内外对Akt蛋白表达和磷酸化水平的影响。方法不同剂量LPS攻击RAW264.7细胞,检测不同时相点细胞Akt蛋白表达和磷酸化水平。小鼠腹腔注射LPS6h后检测肺组织Akt蛋白表达和磷酸化水平。结果不同剂量LPS攻击RAW264.7细胞,细胞Akt蛋白表达没有显著差异,磷酸化水平增加。内毒素血症小鼠肺组织Akt蛋白表达无显著变化,磷酸化水平降低。结论LPS在体内外对Akt蛋白磷酸化作用不同,体外Akt蛋白磷酸化水平增高,但内毒素血症小鼠体内出现Akt蛋白磷酸化水平异常降低。
Objective To explore the effects of LPS on Akt expression and phosphorylation. Methods RAW264.7 cells were attacked with LPS. The Akt protein expression and phosphorylation level were detected by Western blot. The Akt protein phosphorylation level in the lung tissue of mice was detected by Western blot 6 hours after LPS attack. Results The Akt protein expression of RAW264.7 cells showed no obvious difference after attacks of different - dose of LPS, but their phosphorylation level were increased significantly. Whereas the Akt protein phosphorylation level decreased in the lung tissue of mice 6 hours after LPS attack. Conclusion The Akt protein phosphorylation level of Akt protein were different after LPS attack in vitro and in vivo. The Akt protein phosphorylation were increased obviously in vitro, but decreased in a murine model of endotoxemia.
